Hello Stident
Separation and isolation of DNA fragment are follows;
- Cutting of DNA by restriction endonuclease results in DNA fragments
- The fragments are separated by gel electrophoresis
- the commonly used gel is Agarose gel, anatural polymer found in sea weeds.
- Agarose gel provides a sieving effect means separate according to their size,smaller fragment,farther it moves.
- After that these are stained withEthidium Bromide followed by UV exposure.
- Finally Elution occurs in which separated bands of DNA are cut from the agarose gel.
