Separation and isolation of DNA fragment are follows;

  • Cutting of DNA by restriction endonuclease results in DNA fragments
  • The fragments are separated by gel electrophoresis
  • the commonly used gel is Agarose gel, anatural polymer found in sea weeds.
  • Agarose gel provides a sieving effect means separate according to their size,smaller fragment,farther it moves.
  • After that these are stained withEthidium Bromide followed by UV exposure.
  • Finally Elution occurs in which separated bands of DNA are cut from the agarose gel.
